O ácido lisofosfatídico (LPA) é un derivado de fosfolípido que actúa na célula como molécula sinalizadora.[1][2] Está formado por un glicerol esterificado por un fosfato e un ácido graxo (só un, xa que o outro OH do glicerol queda libre)[3].

Función

O ácido lisofosfatídico actúa como un potente mitóxeno debido á súa activación de tres receptores acoplados á proteína G de alta afinidade chamados receptores de lisofosfolípidos 1, 2 e 3 ou LPA1, LPA2 e LPA3 (tamén coñecidos como EDG2, EDG4, e EDG7). Ademais, descubríronse recentemente novos receptores como LPA4 (p2y9/GPR23), LPA5 (GPR92) e LPA6 (GPR87).

Importancia clínica

Debido á súa capacidade para estimular a proliferación celular, a sinalización aberrante con ácido lisofosfatídico foi asociada co cáncer por numerosas vías [4]. A desregulación da autotaxina ou dos receptores do ácido lisofosfatídico pode orixinar hiperproliferación, o cal pode contribuír á oncoxénese e metástase.

O ácido lisofosfatídico pode ser a causa de proídos [5] en persoas con doenzas colestáticas (co fluxo biliar anormal).

Activación da GTPase

Despois da activación do receptor do ácido lisofosfatídico, a pequena GTPase Rho pode ser activada, activando seguidamente á Rho quinase. Isto pode levar á formación de fibras de estrés e á migración celular por medio da inhibición da miosina de cadea lixeira fosfatase.

Metabolismo

Hai varias rutas potenciais parta a biosíntese de ácido lisofosfatídico, pero a mellor caracterizada é aquela na que actúa a lisofosfolipase D chamada autotaxina, a cal elimina o grupo colina da lsofosfatidilcolina.

O ácido lisofosfatídico é tamén un intermediario na síntese de ácido fosfatídico (que ten dous ácidos graxos).
