Lot-et-Garonne on departementti Lounais-Ranskassa Uuden-Akvitanian hallintoalueella. Se on saanut nimensä Garonne- ja Lotjokien mukaan. Departementti koostuu neljästä arrondissementista, 40 kantonista ja 319 kunnasta. Sen pinta-ala on 5 361 neliökilometriä ja siellä on 322 283[1] asukasta (vuonna 2006).

Lot-et-Garonne on yksi Ranskan 83:sta alkuperäisestä departementista, jotka perustettiin Ranskan vallankumouksen jälkeen vuonna 1790.
