الفانكوميسين (بالإنجليزية: Vancomycin)‏ هو مضاد حيوي يستعمل لمكافحة العدوى النّاتجة عن البكتيريا إيجابية الغرام. تصنف على أنها من المضادات ذات آخر لجوء، بمعنى أنه يتم استخدامها كآخر حل عند فشل المضادات الحيوية الأخرى، لكن مع ظهور أشكال حيوية جديدة مضادة للفانكوميسين، بدأ هذا المضاد يفقد دوره بأنه مضاد ذو آخر لجؤ لصالح أنواع جديدة من المضادات مثل لينزوليد ودابتوميسين.

لمحة تاريخية

تم عزل الفانكوميسين لأول مرة بواسطة إدموند كورنفيلد (يعمل لدى شركة إيلي ليلي) من عينة تربة جمعت من داخل أدغال بورنيو أثناء حملة تبشيرية.[5] سمي الكائن المنتج له لاحقاً Amycolatopsis orientalis.[6] دواعي استعمال عقار الفانكوميسين كانت أصلاً لمعالجة العنقوديات الذهبية المقاومة للبنسلين.[6][7]

المركب أطلق عليه مبدئياً بمركب 05865، ثم أطلق عليه فيما بعد الاسم الجنيس فانكوميسين (مشتقة من الكلمة الإنجليزية vanquish وتعني يقهر أو يهزم).[6] من الفوائد التي ظهرت بشكل أن العنقوديات لم تطور مقاومات ذات شأن بالرغم من سلسلة المزارع التي تحتوي على الفانكوميسين. إن تطور العنقوديات السريع لمقاومة البنسلين أدى إلى سرعة إقرار المركب الجديد بواسطة إدارة الأغذية والأدوية الأمريكية في 1958م. قامت شركة إيلي ليلي بتسويق المركب تحت الاسم التجاري فانكوسين.[7]

لم يصبح فانكوميسين الخيار الأول لمعالجة العنقوديات الذهبية لعدة أسباب:

  1. إن العقار يعطى وريدياً، لأنه غير قابل للامتصاص عن طريق الفم
  2. مشتقات جديدة للبنسلين أقوى تم تطويرها فيما بعد مثل المثسلين (وما تبعه كالنافسلين الكلوكسلين).
  3. التجارب الأولية للصيغ الأولية من مركب الفانكوميسين والتي لم تكن نقية (وحل الميسسبي) والتي وجدت لها تأثيرات سُمّّّّّية على الأذن والكلى، هذه الاكتشافات صنفت الفانكوميسين على أنه عقار ذو آخر لجوء

في سنة 2004، رخصت شركة ايلي ليلي الغانكوسين لصالح صيدليات فايروفارما في الولايات المتحدة الأمريكية، صيدليات فلين فارما في المملكة المتحدة وصيدليات آسبن فارمكير في أستراليا. انتهى تسجيل براءة اختراع المركب في بدايات الثمانينات (في السنوات التي تلي 1980)وأصبحت صيغ شائعة من العقار متاحة تحت أسماء تجارية أخرى.
