Juan Augusto de Anhalt-Zerbst (Zerbst, 29 de julio de 1677-ibidem, 7 de noviembre de 1742) fue un príncipe alemán de la Casa de Ascania y gobernante del principado de Anhalt-Zerbst.
Era el hijo mayor del príncipe Carlos Guillermo de Anhalt-Zerbst y de su esposa, Sofía, hija del duque Augusto de Sajonia-Weissenfels.
En 1718, tras la muerte de su padre, Juan Augusto lo sucedió como príncipe de Anhalt-Zerbst.
Juan Augusto contrajo matrimonio con Federica (Gotha, 24 de marzo de 1675-Karlsbad, 28 de mayo de 1709) hija del duque Federico I de Sajonia-Gotha-Altemburgo, el 25 de mayo de 1702 en Zerbst. No tuvieron hijos. Contrajo matrimonio por segunda vez con Eduviges Federica (Weiltingen, 18 de octubre de 1691-Zerbst, 14 de agosto de 1752) hija del duque Federico Fernando de Wurtemberg-Weiltingen, el 8 de octubre de 1715 en Zerbst. Esta unión tampoco tuvo descendencia.
En tanto que Juan Augusto murió sin descendientes, la línea mayor de Anhalt-Zerbst quedó extinta. A su muerte, fue sucedido por sus primos lejanos, los príncipes de Anhalt-Dornburg.
