Dichlorphenolindophenol (DCPIP, DCIP or DPIP) ist eine chemische Verbindung, die als Redoxfarbstoff verwendet wird. Oxidiert ist DCPIP blau mit einer maximalen Absorption bei 600 nm; im reduzierten Zustand ist DCPIP farblos.

Verwendung

DCPIP kann zur Messung der Photosyntheserate verwendet werden. Es gehört zur Familie der Hill-Reagenzien. Bei Lichteinwirkung in einem photosynthetischen System wird der Farbstoff durch chemische Reduktion entfärbt. DCPIP hat eine höhere Elektronenaffinität als Ferredoxin und die photosynthetische Elektronentransportkette kann DCPIP als Ersatz für NADP+ reduzieren, das normalerweise der letzte Elektronenträger bei der Photosynthese ist. Wenn DCPIP reduziert und damit farblos wird, kann die daraus resultierende Erhöhung der Lichtdurchlässigkeit mit einem Spektralphotometer gemessen werden.


Die Reduktion von DCPIP

DCPIP kann auch als Indikator für Vitamin C verwendet werden.[2][3] In Gegenwart von Vitamin C, einem guten Reduktionsmittel, wird der blaue Farbstoff, der sich unter sauren Bedingungen Pink verfärbt, durch Ascorbinsäure zu einer farblosen Verbindung reduziert. Diese Reaktion ist eine Redoxreaktion: Vitamin C (Ascorbinsäure) wird zu Dehydroascorbinsäure oxidiert, und DCPIP wird zur farblosen Verbindung DCPIPH2 reduziert.

Bei dieser Titration stehen nach Verbrauch der gesamten Ascorbinsäure in der Lösung keine Elektronen mehr zur Reduktion von DCPIPH zur Verfügung, und die Lösung bleibt aufgrund des DCPIPH rosa. Der Endpunkt ist eine Rosafärbung, die 10 Sekunden oder länger anhält, wenn nicht genügend Ascorbinsäure vorhanden ist, um das gesamte DCPIPH zu reduzieren. Pharmakologische Experimente deuten darauf hin, dass DCPIP als prooxidatives Chemotherapeutikum gegen menschliche Krebszellen in einem Tiermodell des menschlichen Melanoms wirken könnte; der DCPIP-induzierte Krebszelltod erfolgt durch Mangel an intrazellulärem Glutathion und Hochregulierung von oxidativem Stress.[4]
