Grand Prix sepeda motor Italia 2016 adalah putaran keenam dari MotoGP musim 2016. Itu diadakan di Mugello Circuit di Scarperia pada tanggal 22 Mei 2016. Balapan MotoGP diselesaikan hanya dengan 0,019 detik saat Jorge Lorenzo keluar dari slipstream Marc Márquez di lintasan lurus terakhir di lap terakhir setelah duel dramatis antara keduanya, sementara polesitter Valentino Rossi kehilangan kesempatan untuk menantang kemenangan kandang karena kerusakan mesin.[4] Balapan ini menandai kegagalan motor pertama Rossi sejak Grand Prix San Marino 2007.

Balapan ini merupakan balapan terakhir bagi Luis Salom sebelum ia mengalami cedera parah setelah mengalami kecelakaan saat sesi latihan di Grand Prix selanjutnya di Barcelona.

Di kelas Moto3, Fabio Di Giannantonio meraih podium pertamanya dalam start ke-7.
