    • 所要時間約42分。運賃は大人820円、子供410円。平日朝に1便のみ片道運行される。便によっては下田水港を経由しない。

特急便

下記の2便は瀬戸内運輸の担当便。特急便は下田水港・平原・大島営業所(宮窪)・多々羅の里・中央橋・教善寺・宮浦農協には停車しない[2]。

今治北IC - 大三島IC間はしまなみ海道経由。ただし大島南IC - 大島北IC間は国道317号を走行する。しまなみ海道経由のためシートベルト付きの車両で運行され着用が義務付けられている。瀬戸内運輸の特急バスも同様。立ち席を認めていないため満席時は乗車できない（座席定員制）。瀬戸内運輸便と共に全席禁煙となっている。

  • 特急 松山市駅 - 今治 - 大三島宮浦港
    • 松山市駅 - 大街道 - 奥道後 - 玉川支所 - 馬越 - 今治駅前 - 今治バスセンター - 今治桟橋 - 今治バスセンター(折り返しの都合上2回経由) - 今治県病院 - 馬島BS - 吉海支所 - 伯方島BS - 大三島宮浦港
    • 松山まで行く便で、急行便とは一部停車停留所が異なる。主要停留所のみ記載。
    • 松山発着便はかつて伊予鉄バスも運行していたが撤退した。また当初の菊間町経由から、のちに玉川経由へ経路変更された。
  • 特急 今治桟橋 - 伯方島
    • 伯方島BS - 石文化運動公園 - 吉海支所前 - 亀山 - 馬島BS - 今治県病院 - 今治駅前 - 今治バスセンター - 今治桟橋

廃止路線

  • 瀬戸田 - 宮浦港
    • 専用のハイデッカー車を導入したが開業当初から不振で廃止。専用車両は今治線に転用された。
  • 宮浦港 - 盛港 - 忠海港 - 三原市（広島県）
    • 瀬戸田線と同様に開業当初から不振で廃止。なおこの路線は全国的にも珍しく、盛港 - 忠海港（広島県竹原市）間はバス自体をフェリーに積み込み運行していた。
  • 木浦港 - 瀬戸浜循環 - 木浦港（北回り・南回り）
  • 木浦港 - 下田水港
  • 田浦 - 亀山 - 下田水港
  • 宮浦港 - 肥海 - 盛港
  • 宮浦港 - 森側 - 盛港
    • この路線の廃止により、盛方面のバス路線は消滅した。
  • 宮浦港 - 大三島BS - 伯方島BS - 下田水港
    • この路線は1988年の伯方・大島大橋開通後に開設されて以来、約20年にわたり運行されてきたが、1999年の来島海峡大橋開通で開設された、大三島 - 今治線に客足が流れ利用者が激減。減便を繰り返しつつ路線維持してきたが、輸送旅客数の増加が見込めず、国・愛媛県・今治市から補助金を受けられる基準を満たす可能性が低くなったことから、2007年9月末をもって廃止された。
  • 下田水港 - 椋名 - 吉海支所前 - 田浦[3]
  • 宮浦港 - 井口港 - 大三島BS - 瀬戸 - 出走[4]
