Alben William Barkley lahir di dekat Lowes, Graves County, Kentucky, pada 24 November 1877. Orangtuanya, John Wilson Barkley dan Electra Eliza Smith adalah petani yang religius. Nama asli Alben ialah Willie Alben Barkley. Ia masuk Marvin College, Clinton, Kentucky di mana ia terkenal piawai dalam pidato dan debat. Barkley lulus pada tahun 1897, lalu melanjutkan studi di Emory College, Oxford, dan lulus tahun 1900. Di sini ia menjadi anggota klub Delta Tau Delta. Barkley terpilih sebagai Wakil Presiden ke-35, mendampingi Presiden Harry Truman, mulai Januari 1949 sampai Januari 1953. Saat dilantik ia berusia 71 tahun dan merupakan Wapres tertua dalam sejarah AS. Barkley juga pernah dianugerahi Medali Emas Kongres tahun 1949.

Barkley kembali ke Senat AS pada tahun 1955 sampai akhir hayatnya. Suami dari Jane Hadley ini meninggal dunia saat berpidato di Washington and Lee University, di kota Lexington, Virginia, pada tanggal 30 April 1956. Jenazahnya dimakamkan di Mount Kenton cemetery, dekat Danau Oak Road, Paducah, Kentucky.
