Die Fußball-Stadtliga Berlin 1959 war die achte Spielzeit der vom Bezirksfachausschuss (BFA) Fußball Berlin (Ost) durchgeführten Bezirksliga Berlin und die Dritte unter der Bezeichnung Stadtliga. Sie war die höchste Spielklasse in Berlin (Ost) und die vierthöchste im Ligasystem auf dem Gebiet des DFV.

Die BSG Tiefbau Berlin sicherte sich den Stadtmeistertitel auf Grund des besseren Torverhältnisses gegenüber der BSG Motor Köpenick und stieg in die übergeordnete II. DDR-Liga auf. Köpenick verpasste den Titel am letzten Spieltag, durch ein 0:0 im Heimspiel gegen Johannisthal.

In die untergeordnete Stadtklasse stieg nach sechsjähriger Zugehörigkeit die SG Sportfreunde Johannisthal ab. Im Gegenzug stiegen zur Folgesaison die beiden Erstplatzierten die SG Fortuna Biesdorf als Neuling und nach vierjähriger Abwesenheit die BSG Lok Schöneweide in die Stadtliga auf. Aus der II. DDR-Liga kam noch die BSG Berliner Verkehrsbetriebe dazu.
