At the time of the 2006 National Census, Shamkan's population was 2,102 in 541 households, when it was a village in Shamkan Rural District of the former Sheshtamad District of Sabzevar County.[6] The following census in 2011 counted 2,439 people in 710 households.[7] The 2016 census measured the population of the village as 1,882 people in 581 households. It was the most populous village in its rural district.[2]
In 2019, the district was separated from the county in the establishment of Sheshtamad County, and the rural district was transferred to the new Shamkan District.[4] Shamkan was elevated to the status of a city in 2022.[8]
