Penghambat pompa proton (Bahasa Inggris: Proton-pump inhibitors, disingkat PPIs atau PPI) adalah kelompok obat yang beraksi utama dalam pengurangan produksi asam lambung. Dalam kelas obat, tidak ada bukti jelas bahwa satu agen bekerja lebih baik daripada yang lain.[1]

PPI merupakan obat paling ampuh dalam menekan sekresi asam lambung .[2] Kelompok obat lain yang bekerja untuk menekan produksi asam lambung yaitu antagonis reseptor H2.

PPI merupakan salah obat yang paling banyak dijual di dunia, dan yang pertama, omeprazol, adalah Daftar Obat Esensial model WHO.[3] Biaya antara agen yang berbeda bervariasi secara signifikan.
